Male C57BL/6 mice of approx. age of 6 months, and on high-fat diet for approx. 3 months were injected subcutaneously with 75,000 cells of the KPN1.1 mouse lung carcinoma cell-line. Starting after about a week, mice received daily injections of either phosphate-buffered saline (PBS) or liraglutide (0.2 ug per g body weight, in PBS) till the end of experiment, when lungs and tumors were collected and flash-frozen for storage at -80 deg. C. Tissues of 5 mice of each treatment group were homogeneized for RNA isolation, in a single batch, with Qiagen MiRNeasy Micro Kit followed by quantification with Thermo HS RNA Qubit and Agilent HS RNA ScreenTape assays. Sequencing libraries were prepared using 500 ng RNA with KAPA RNA HyperPrep with RiboErase kit and 12 PCR cycles. Sequencing of all 20 libraries (200 pmoles each) was done in the same run on NovaSeq 6000 with S2 cell and Reagent Kit v1.5 (200 cycles) to obtain 101 b paired-end reads.
