Cisplatin resistance in A2780 ovarian cancer cells was induced by treatment with gradually increasing concentrations of cisplatin. Transcriptomic differences between cisplatin-resistant and parental A2780 cells were analyzed using RNA sequencing (RNA-Seq), while changes in RNA 5-methylcytosine (m5C) methylation levels were assessed using bisulfite sequencing (BIS-Seq). To investigate the role of the m5C reader protein ALYREF, its expression was knocked down in cisplatin-resistant cells, followed by RNA-Seq to examine resulting gene expression changes. ALYREF-bound RNAs were identified by RNA immunoprecipitation sequencing (RIP-Seq).
