CTCF deficiency in CD8 homeostatic proliferation [RNA-seq]

An intricate interplay among Tcf1/Lef1 and CTCF that orchestrates genomic architecture to coordinately promote CD8+ T cell homeostatic proliferation Splenocytes were harvested from hCD2-Cre, Rosa26-lsl-GFP, wild-type, Tcf1/Lef1-double floxed, or Ctcf-floxed mice, and enriched for naive CD8+ T cells by depleting non-T lineage cells, CD4+ and CD44 high cells. The enirched cells were labeled with CTV and transferred into Rag1 KO recipients. 72 hrs later, CTV+TCRbeta+GFP+CD8+ T cells were sorted from the recipient spleens and total RNA extracted.
