The goal of this study is to compare the gene expression profiles of two populations of human monocyte-derived macrophages (HMDMs) that were subjected to differentiation in either of two colony-stimulating factors: M-CSF or GM-CSF. Specifically, we sought to characterize the transcriptional responses of M-CSF and GM-CSF macrophages to the molecules, adenosine and prostaglandin E2 (PGE2) coupled with LPS stimulation. We identified gene expression that is enriched in HMDMs stimulated with LPS+Adenosine and LPS+PGE2 relative to LPS alone, as these two molecules have been known to alter macrophage inflammatory responses. This work demonstrates that M-CSF and GM-CSF human macrophages have distinct transcriptional alterations following stimulation in the presence of Adenosine and PGE2.
