We placed immotile variants of the soil bacterium Pseudomonas fluorescens Pf0-1 (Pf0-1(delta)fleQ; Pf0-2x) under selection for recovered flagella-mediated motility using soft agar as a directional selection medium. We observed that typically a single de novo mutation would arise within 72-144 hours and restore the motility phenotype. We sent a subset of evolved lines from this experiment for whole-genome sequencing to identify the phenotype-causing mutation and screen for secondary, background and hitchhiking mutations. One mutant line was identified to harbour two nucleotide polymorphisms within the glnA locus (glnA A938C and glnA G943A), which is a member of the nitrogen regulatory pathway associated with recovery of the flagella-mediated motility phenotype.
