Environmental DNA 120 samples from Dutch freshwater lakes was metabarcoded using general COI primers. The lakes were sampled on a spatial and temporal scale, to investigate the patterns of eDNA in space and time. The goal of the study is to analyze the impact of three levels of levels of replication (spatial and temporal sampling, and PCR replication), allowing for better optimization of sampling strategies for environmental DNA monitoring.
