Endocytosis is a cellular process of internalizing materials surrounded by an area of the cell membrane. This thesis addresses the dynamic regulation of endocytosis at several stages of maturation, utilizing advanced microscopic techniques. This includes understanding the membrane ruffling induced by Rac1 to drive internalization through macropinocytosis, molecular mechanisms underlying the trafficking of activated EGFR to the perinuclear region, role of PI(3)P in sorting of cargoes from early endosomes. Further addresses the mechanisms of intracellular transport in the disease context such as Amyloid-β trafficking in Alzheimer’s disease and role of βIII-tubulin in regulating the mitochondrial dynamics.
