Epigenetic profiling in 4 cohorts of pancreatic cancer patients revealed reduced expression of the chromatin factor inhibitor of growth family member 3 (ING3). Furthermore, using mass spectrometry analysis we demonstrated that the linker Histone 1.2 (H1.2) is a direct binding partner of ING3. Thus, we reasoned to explore the Genome binding/occupancy profiling of H1.2 upon ING3 depletion by performing the Illumina NextSeq500 using single-end 75bp protocol. Chromatin fragments, derived from siControl and sING3 treated cells, were immunoprecipitated with antibody against Anti-Histone H1.2. After purification of DNA from cell lysate by functionalized magnetic particles, libraries for next generation sequencing were prepared and analyzed using standard procedures.
