Single-Cell Profiling of Neutrophil Gene Expression in the Mouse Colon following TNBS-induced Colitis

Neutrophil infiltration is hallmark of chemically induced colitis and inflammatory bowel disease (IBD). We used single-cell RNA sequencing to analyze heterogenity in infiltrated colonic neutrophil population in presence and absence of microbiota, or when DUOX2 in inactivated in neutrophils. Mice were treated with 2.5% TNBS intrarectally, and colon was collected after 48 h (day2). Viable neutrophils were purified using FACS (CD11b+, Ly6G+, DRAQ7-) followed by scRNA analysis.
