Oxygen is toxic across all three domains of life. Yet, the molecular mechanisms of hyperoxia pathology remain largely unknown. The study set out to uncover mechanisms of oxygen toxicity identified using genome-wide CRISPR/Cas9 screen. We further validated the finding in a murine model of hyperoxia-induced pulmonary injury. This work has important implications for hyperoxia pathologies, including bronchopulmonary dysplasia, ischemia-reperfusion injury, aging, and mitochondrial disorders.
