We report that the use of high throughput RNA sequencing in C2C12 myotubes induced myotobule hypertrophy and promoted ECM remodeling through alterations in collagen synthesis and turnover. Transcripts for IGF-binding proteins and Fibromodulin suggest increases in growth and a suppression of myostatin. Overall design: Examination of fully mature myotubes that were treated with either 4% horse serum differentiation media (DMEM, 4%HS, Pen/Strep) or 4% STEM cocktail media (DMEM, 4% STEM, Pen/Strep). Tubes were differentiated in 2% HS differentiation media for 5 days. At day 6 each respective treatment was applied to cells for a duration of 24 hours. After 24 hours cells were collected and RNA isolated and given to the University of Utah Genomics Core for RNA sequencing.
