In order to identify genomic regions which tolerate loss-of-function mutations among a collection of 112 Saccharomyces cerevisiae isolates, a saturated transposon mutagenesis approach was performed to randomly insert a transposon in the genome. For each sample, the genomic DNA of the pool of cells which survived to the insertion event was extracted and the transposon insertion region was sequenced (Illumina HiSeq or DNBseq technologies). A mapping of the precise genomic insertion site and the comparison of the transposon insertion profiles between the different genetic backgrounds highlighted genes or even regions with conditional loss-of-function phenotypes.
