The emergence of multi-drug resistant tuberculosis threatens global efforts for disease control of tuberculosis. To identify novel genes associated with isoniazid (INH) resistance, whole-genome sequencing of 12 INH-resistant and 17 INH-susceptible Mycobacterium tuberculosis clinical isolates was performed. Sanger sequencing of 58 selected genes in additional strains revealed 104 novel mutations associated with INH-resistance. Unique spoligotype-specific mutational signatures suggest that mutations in multiple genes occur in subsets of INH-resistant isolates.
