Intrinsic and Extrinsic Constraints on Mammary Gland Development Induced by Pubertal High-Fat Diet

This dataset accompanies the study “Intrinsic and Extrinsic Constraints on Mammary Gland Development Induced by Pubertal High-Fat Diet.” The study investigates how pubertal high-fat diet (HFD) exposure alters mammary gland development using mouse mammary gland single-nucleus RNA sequencing and integrated single-cell transcriptomic reference analyses. The deposited data include two related resources: the normal diet (ND) dataset and the high-fat diet (HFD) dataset. The ND and HFD datasets contain single-nucleus RNA-seq profiles generated from mammary gland tissues of female C57BL/6N mice fed either a 10% kcal fat normal diet or a 60% kcal fat high-fat diet from 3 weeks to 10 weeks of age. These data were used to characterize diet-associated changes in mammary epithelial, stromal, adipocyte, endothelial, and immune compartments, including alterations in epithelial abundance, luminal growth signaling, extracellular matrix remodeling, adipocyte-state transitions, and macrophage–epithelial communication. The Reference dataset integrates in-house ND mammary gland data with publicly available prepubertal and pubertal mouse mammary gland single-cell RNA-seq datasets with the scripts described in our manuscript.
