Burkholderia multivorans is a member of the Burkholderia cepacia complex (Bcc) whose members are inherently resistant to many antibiotics and can cause chronic lung infections in patients with cystic fibrosis (CF). A possible treatment for chronic infections arises from the existence of collateral sensitivity (CS) acquired resistance to a treatment antibiotic results in a decreased resistance to a non-treatment antibiotic. Previously we have shown that sequential drug therapy in the majority of B. multivorans isolates tested can induce CS responses to, at minimum, one class of antibiotics. However, a small proportion of the CS isolates that initially responded to sequential drug treatment developed extensive drug resistance (XDR) to at least four different classes of antibiotics. Here we show that upon acquisition of resistance to a fifth antibiotic, the combination drug sulfamethoxazole-trimethoprim, observed increased sensitivity to all four classes of antibiotics. Identified mutational events of XDR parent strains and CS progeny.
