Nardostachys jatamansi is a Himalayan medicinal plant, known for its excellent antioxidant and metabolic activity. Two different gold nanoparticles with distinct charge on the surface (positive and negative) modulate growth, photosynthetic activity and antioxidant potential of the plant. To decipher the transcripts and pathways underlying gold nano elicitation of N. jatamansi, transcriptome of in-vitro shoots subjected to control (NC), negative charged AuNPs (NT1) and positive charged AuNPs (NT2) was sequenced. The study generated total 217.1 million clean reads that identified 104, 1255, and 660 significant differentially expressed genes (DEGs) in NC vs NT1, NC vs NT2 and NT1 vs NT2 libraries, respectively. The Gene ontology, KEGG pathway, DEGs and transcription factors were found to be associated with plant growth, antioxidant activity, hormone metabolism, cellular metabolism and abiotic stress resistance. Altogether, the study provides insights into the transcriptomic response of N. jatamansi during differentially charged gold nanoparticles elicitation and concluded that CTAB coated (positive charged) gold nanoparticles are more efficient than citrate coated (negative charged) gold nanoparticles.
