Single-cell transcriptome analysis revealed an enrichment of Fos+ MPP subpopulations in mouse bone marrow under hypoxic conditions. To validate the biological characteristics and hypoxia stress response of the Fos+ MPP subpopulation, we used Fos-2A-tdTomato mice to establish a HAPC model. We then sorted Fos+ and Fos- MPP cells for low-input DNA methylation sequencing analysis. The DNA methylation experiment consisted of six groups: normoxic control, hypoxia for 2 weeks, hypoxia for 4 weeks, hypoxia for 12 weeks, hypoxia for 12 weeks followed by normoxic recovery for 5 weeks, and hypoxia for 12 weeks followed by normoxic recovery for 5 weeks and re-exposure to hypoxia. For each group, 3-4 mice were used for DNA methylation sequencing. Fos+ and Fos- MPP cells were sorted for low-input DNA methylation sequencing.
