Whole-genome sequencing of Ligilactobacillus salivarius GHL02 derived from poultry lungs

This study aimed to screen poultry lung-derived Lactobacillus strains with effective antibacterial properties. Bronchoalveolar lavage fluid (BALF) was collected from high-performance broiler chickens (43-day-old male chickens weighing 3.3-3.6 kg, n=10) sourced from a large-scale commercial broiler farm, and pooled. The pooled BALF was then enriched in sterile MRS broth followed by plating, and also directly plated onto MRS agar for single colony isolation and purification (incubated at 37 C for 24 h). Subsequent analyses included morphological identification, antibacterial activity assessment, antibiotic susceptibility testing, acid-producing capacity evaluation, and acid and salt tolerance tests. A strain exhibiting superior performance was selected and designated as Ligilactobacillus salivarius GHL02. Whole-genome sequencing of this strain was performed using a combination of second-generation Illumina sequencing and third-generation Nanopore sequencing.
