This study developed a concise and accessible procedure for mapping cotranscriptional RNA folding pathways using high-throughput RNA structure probing. The BioSamples associated with this BioProject contain data collected during the initial validation and application of the method, called Transcription Elongation Complex structure probing - multilength (TECprobe-ML). In this study, the folding pathways of a C. beijerinckii ZTP riboswitch, a B. cereus fluoride riboswitch, and a C. bacterium ppGpp riboswitch were characterized. These data establish the accuracy of the method, which can be applied generally to understand the mechanisms and determinants of cotranscriptional RNA structure formation.
