To investigate transcriptional role of TgAP2XII-1 on type I Toxoplasma gondii (RH?hxgprt Tir1), an auxin inducible degron system was introduced to control the protein expression of TgAP2XII-1 and the target protein could be totally degradated in 3h after adding IAA. Overall design: Transgenic parasite iKD AP2XII-1-mAID-3HA was treated with IAA for 0h, 12h or 24h, then total RNA were extracted for deep sequencing.
