Effect of the expression of HP1 proteins from different species (mouse, fruit fly, fission yeast) in mouse 3T3 cells

HP1 proteins play a role in heterochromatin regulation and higher-order chromatin organization. The domain structure of HP1 is conserved, but the properties and functions of HP1 homologs from different organisms might vary. To address this issue, we expressed HP1 from different species in mouse 3T3 cells and assessed the impact on gene expression by RNA-seq. Overall design: To test the effect of different HP1 homologs on gene expression, we established stable 3T3 cell lines with GFP-tagged HP1 from fission yeast, fruit fly and mouse under the control of a doxycycline-inducible promoter. We then performed total RNA-seq in doxycycline-induced (GFP-positive during sorting) and uninduced cells.
