In recent years, scRNA-seq studies on the decidua associated with RSA have focused on the developmental defects of stromal cells and the decidual immune microenvironment in RSA patients. Our study aims to delve deeper into the pathogenesis of RSA, while further clarifying the potential mode of action of WJ-MSCs in the treatment of RSA. In conclusion, our study mapped the single cell profiles of the decidua in three groups of mice with normal pregnancy, RSA and WJ-MSCs-treated RSA by single cell RNA sequencing, described the functional characteristics and pseudo-temporal trajectories of each subpopulation of stromal cells, the major cell population of the decidua, further resolved the decidua heterogeneity characteristics, indicated the signalling pathways of cellular interactions imbalance in the decidual microenvironment of RSA, and further illustrated the It further illustrated the interactions between stromal cells and NK cells, and revealed the potential therapeutic mechanism of WJ-MSCs to remodel the subpopulation of RSA molting cells, providing a new idea for understanding the developmental defects of decidua at the maternal-fetal interface of RSA and the therapeutic potential of WJ-MSCs.
