The zBmal2 and zBmal2_mut plasmids were linearized by ApaI and transcribed into mRNAs separately using mMESSAGE mMACHIN SP6 Transcription Kit . The mRNA products were finally diluted to 400 ng/ul for microinjection. The mRNA products from the two treatment groups (zbmal2 and zbmal2_mut) were microinjected into the one-cell-stage wild-type zebrafish embryos using the ASI pressure injection system under a stereomicroscope.
