Spatial immune heterogeneity in mouse tumors

To identify drivers of heterogeneity in the immune tumor microenvironmentupon immunotherapy, mice were subcutaneously inoculated with TC-1/A9 tumorcells and treated with combined immunotherapy consisting of DNA vaccinationand administration of immunostimulatory CpG motifs. Mock-treated miceserved as controls. In tumor sections, 2-3 morphologically separated areaswere localized, and RNA and DNA were isolated after macrodissection of thesample tissue. RNA sequencing was used for gene expression analysis andwhole exome sequencing for mutation identification.
