Gene expression analysis of alveolar macrophages from BCG-vaccinated and CoMtb mice stimulated ex vivo [exvivoRNAseq]

The aim of this study was to measure the response of alveolar macrophages (AMs) from BCG-vaccinated mice and mice with CoMtb to LPS, PAM3CSK4, and Mtb stimulation ex vivo. Mice were vaccinated sub-cutaneously with BCG-Pasteur (1e6 CFU), or inoculated in the dermis of the ear with H37Rv (1e4 CFU) and allowed to rest for 8 weeks. To characterize the macrophage transcriptional response to each stimulus, AMs were isolated by bronchoalveolar lavage followed by plate adherence for 24 hours. AMs were stimulated with either LPS (10 ng/mL), Pam3CSK4 (10 ng/mL), or Mtb-H37Rv for 6 hours prior to RNA isolateion.
