This work describes flow-cytometry based enrichment of human enteric neurons and glia using a combination of cell surface markers (CD56/CD90/CD24) which enable studies of the human ENS. Overall design: Pediatric small intestine and colon was enzymatically digested using Collagenase and Dispase to obtain a single cell suspension. Cells were stained with fluorescent antibodies (CD56/CD90/CD24). Enteric nervous system (ENS) cells were enriched by Fluorecence-activated cell sorting (FACS) according to the CD56+CD90+ gate. Sorted cells from 4 donors were processed with the 10X Gene expression profiling technology.
