Sediment slurry from a hot spring was incubated with a synthetic amino acid with various substrate amendment or headspace changes. Fluorescently labeling the synthetic amino acid facilitated FACS of organisms that were active under the conditions and incorporated the amino acid into newly synthesized proteins. By sorting active cells from the bulk communities, microbial responses to substrate treatment could be determined using 16S rRNA gene sequence analysis.
