Sanger sequencing of PCR products amplified with degenerate oligonucleotide primer (DOP) pair using the genomic DNA extracted from the large cabbage white, Pieris brassicae, as a template. DOP was designed based on sequence information of Bombyx mori and Tribolium castaneum genes. All the 41 sequences obtained were confirmed to be B. mori orthologs. These sequence information was further used to design single tagged site (STS) and the primer pairs were used to select the bacterial artificial chromosome (BAC) clones containing the orthologs. We extracted the BAC-DNA to make probes for BAC-FISH (fluorescence in situ hybridization) mapping in P. brassicae and P. rapae.
