Using HTA 2.0 microarrays, we demonstrate transcriptome changes in ovarian cancer SKOV3 cells induced by hypoxia, mitomycin C and their combined effect with particular emphasis on the effect of oxygen tension modification on mitomycin c activity. Examining differentially expressed genes (DEGs) in these groups. Total RNA of SKOV3 cells untreated/treated with mitomycin C (MMC) in normoxia or hypoxia (4 experimental groups in total- with 4 biological replicates for each group: normoxia control -without treatment = N-C, normoxia with MMC treatment = N-MMC, hypoxia control - without treatment = H-C, hypoxia with MMC treatment = H-MMC) was extracted and amplified 100 ng of RNA per sample were then hybridized on GeneChip Human Transcriptome Array 2.0 (HTA 2.0) (Affymetrix, Thermofischer scientific). Differentially expressed genes (DEGs) in these groups were examined.
