Single-cell analyses were conducted to further elucidate the fibrosis-inhibitory and cardioprotective effects of AZD4547, as well as the impact of cell-cell interactions. To validate these effects in the acute phase, analyses were performed at day 7 left ventricular heart samples from control, PE/Ang II-exposed without treatment, and AZD-administered mice. Overall design: Mouse heart dissection and single-cell preparations were performed according to the protocols provided by 10x Genomics. In brief, hearts were collected from three groups: control, AngII/PE with AZD treatment, and AngII/PE with NS treatment, seven days post pump implantation. The left ventricular (LV) chambers were anatomically dissected, and the tissues were flash-frozen in liquid nitrogen for subsequent analysis.
