We aimed to detect the difference in immune cells after immunization of the MC38 subcutaneous transplantation model with the cancer vaccine Adpgk-NitraTh. NitraTh is a kind of helper CD4 T cell epitope containing p-nitrophenylalanine, which we have confirmed to have great anti-tumor reactivity. We used single-cell sequencing to further explore the mechanisms behind this activity, in comparison with the common helper CD4 T cell epitope PADRE.Overall, we divided the mice of the MC38 subcutaneous transplantation model into three groups, vehicle, Adpgk-PADRE, and Adpgk-NitraTh, using different vaccines to immune them. After sacrificing, we collected the tumor tissues and separated them into single-cell suspensions. Finally, we used CD45+ microbeads to collect immune cells from suspensions and sent them for single-cell sequencing.
