The aim of this study was to estimate the effects of CO2 availability on the community structure and photosynthetic physiology of diatoms in the spring Oyashio water. Water samples were collected from 10 m depth at a station PH3 in the Oyashio region of the western North Pacific on 9 May, during the KT-11-7 cruise of R/V Tansei-Maru (JAMSTEC). A short-term (3 days) on-deck incubation experiment was conducted under different CO2 levels (180, 350, 750, and 1000 ppm), as a part of the Plankton Ecosystem Response to CO2 manipulation Study (PERCOM). The DNA and RNA samples were collected on days 0 and 3. The sequences of diatom-specific rbcL fragments were then obtained using Ion Torrent amplicon sequencing technology.
