The research object of this project is the continuous isolation of Staphylococcus aureus from a patient's body, collected through routine sampling and microbial culture in the Clinical Microbiology Laboratory of the First Affiliated Hospital of Hebei North University according to medical orders. The phenotypic characterization of these strains, such as minimum inhibitory concentration (MIC) determination, pulsed field gel electrophoresis typing, adaptability analysis, biofilm formation ability determination, quantitative determination of golden yellow pigment, mouse virulence determination, etc., was carried out, and the evolutionary trajectory of the strains in vivo was discussed in combination with short reading long and long length whole genome sequencing, comparative genomics analysis, transcriptomics analysis, and RT qPCR verification.
