With the development of tanning technology and biotechnology, enzyme preparations have been widely used in the main processes of tanning. The softening treatment based on acid protease is considered to be an effective way to reduce pollutant emission and collagen loss. However, due to the addition of chromium metal in processes such as chrome tanning, the enzyme activity is inhibited. To develop acid collagenase with high catalytic activity and strong chromium resistance, the enzyme producing microorganism should be mined first. However, the activity of acid collagenase secreted by wild bacteria was low and the tolerance to chromium was low. Therefore, structural modification of acid collagenase is needed.
