Cystine-Based Peptides and Polymer Conjugates as Redox-Sensitive Anion Transporters

This thesis develops redox-sensitive cystine-based peptides and peptide–polymer conjugates for targeted anion transport, with potential use in cancer therapy. Designed to respond to high glutathione levels in triple-negative breast cancer, these peptides, especially bocTrpCystineOMe, efficiently exchange nitrate and chloride ions. Modifying sequences with lysine altered transport selectivity and efficiency, with some variants enabling hydroxide transport. Polymer conjugation via RAFT synthesis improved stability, membrane binding, and performance at very low concentrations. These systems mimic natural ion channels, offering tunable, cancer-selective ion transport. Future work will enhance stability, lower effective doses, and assess therapeutic potential in advanced models.
