We describe here how inflammation-mediated depletion of miR-126 in endothelial cells (ECs) of aging bone marrow niche contributes to vascular remodeling and reduced supply of EC miR-126 to hematopoietic stem cells (HSCs) and causes age-related decrease of HSC self-renewal capacity. Overall design: RNA-seq were performed with lineage-Sca-1+c-Kit+CD150+CD48- hematopoietic stem cells (HSCs) sorted from 2 month-old young or 20 month-old aged mice (n=3 young HSC samples were pooled from 30 young mice; n=3 aged HSC samples were pooled from 4 aged mice).
