Liposarcoma (LPS) is the most common soft-tissue sarcoma in adults, and well-differentiated LPS (WDLPS) and dedifferentiated LPS (DDLPS) are the most frequent subtypes. These LPSs are considered to develop due to disturbances in the process of adipogenic differentiation of mesenchymal stem cells. However, the molecular mechanism of the disturbances remains unclear. Here, we aimed to identify the mechanism and take its advantage in therapy. Genomic DNA from peripheral blood mononuclear cells, the cell lines from liposarcomas (93T449, 94T778, LP6, and LPS12), and dedifferentiated liposarcoma cell lines (LP6 and LPS12) treated with 5-aza-dC and rosiglitazone were extracted. After bisulfite conversion, DNA was hybridized to the Infinium Human MethylationEPIC BeadChip v1.0 (PBMC and cell lines) or Infinium Human MethylationEPIC BeadChip v2.0 (LP6 and LPS12 treated with mock, 5-aza-dC, and 5-aza-dC and rosiglitazone).
