Over the past several years, replacement chemicals for bisphenol A have been manufactured and used as additives in sportswear, plastic bottles, food packaging, and in other products. More recently, replacement products for BPA are under investigation for toxicity, but there are little data on the potential for neurotoxicity. To address this gap, differentiated human SH-SY5Y cells were treated with a range of concentrations of BPS and BPF to better understand mechanisms of toxicity. Cell culture flasks were exposed to DMSO, or one dose of either BPS or BPF. Cells were exposed in T75 flasks by removing all of the media and replacing it with 8 mL of differentiation media (DMEM:F12 containing 1% FBS, 1% antibiotic/antimycotic solution, and 10 µM retinoic acid) every 48 hours. On the 6th day, cells were exposed to media, 0.1 nM BPS, or 0.1 nM BPF (n=5-6) for 48 hours. Media was then removed and the remaining cells were trizoled and stored at -80°C for further processing.
