Briefly, lung ECs were isolated from EC-specific TMEM215 knockout mice and ctrl mice, and lysed with the TRIzol reagent (Invitrogen). Total RNA was extracted and evaluated for quality by an Agilent 2100 Bioanalyzer (Agilent Technologies, Palo Alto, CA) and RNase free agarose gel electrophoresis. Then, mRNA was enriched, fragmented and reverse-transcribed into complementary DNA (cDNA) with random primers. cDNA fragments were sequenced with the Illumina NovaSeq 6000.
