Ecological Adaptability and Response Mechanism to Salt Stress of Wild Apocynum venetum

We selected wild A. venetum of four distributed regions (jinta county, minqin county, hangjinqi and baicheng) as the test materials in the study. Based on the study of community composition, population genetic diversity and soil environmental properties of wild A. venetum, physiological characteristics, transcriptomics and proteomics analysis of A. venetum under salt stress, and key regulatory proteins and genes of A. venetum salt tolerance were measured and identified to reveal the ecological adaptability of wild A. venetum and underlying mechanisms in response to salt stress. The study is beneficial to promote the protection and rational development and utilization of germplasm resources of wild A. venetum, and is of great significance to the development of national pharmacology and the construction of ecological civilization in China. Overall design: In this study, 18 well-growing wild P. apocinae seedlings were selected and divided into two groups of salt stress treatment group and control group, each treatment was repeated 3 times. 300 mmol/L was selected as the stress concentration for transcriptome test. The test group of P. apocynum plants were carefully washed in pure water, and then transferred to the configured stress nutrient solution (containing 300 mmol/L NaCl). The control plants were washed and transferred to a new nacl-free nutrient solution. At 0 h, 2 h, 6 h, 12 h, 24 h and 48 h stress, Apocynum japonicum leaves were taken and quickly put into the frozen tube for liquid nitrogen deep freezing, and then the samples were stored in the refrigerator at -80? for reserve.
