The aim of this project was to determine the distribution of Phacoides pectinatus, a lucinid bivalve, to assess the diversity of their (endo)symbiont community, and charcterize the free-living bacterial diversity from pore waters and sediments associated with Rhizophora mangle (red mangrove). Sampling took place in July 2014 at Wildcat Cove near Fort Pierce, Florida. Approximately 78% of the live P. pectinatus hosts were discovered within close proximity (i.e., within 1-2 m) to the mangrove canopy, and at depths up to 30 cm in the sediment. Total nucleic acids were extracted from sediment (42 samples), water (9 samples), and the P. pectinatus’ gills and feet (27 samples). DNA was extracted from sediment using MoBioO PowerSoil DNA isolation kits, from filtered pore and ocean water using MoBio PowerWater DNA isolation kits, and from lucinid tissues using Qiagen DNeasy Blood & Tissue kits. From extracted DNA, the V1-V3 region of the bacterial 16S rRNA gene was amplified and single-direction reads were generated using multiplexed, bar-coded primers for pyrosequencing.
