Differentiation of brown adipocytes is a crucial process for adaptive thermogenesis, which is stimulated by various factors. We found robust browning of inguinal white adipose tissue in UCP1/ApoE-DKO mice, but not in ApoE-KO mice, under high-fat diet condition. We used microarray to determine the genes specifically regulated in the browning white adipose tissue in UCP1/ApoE-DKO mice. Male 9-week-old littermates of ApoE-KO and UCP1/ApoE-DKO mice were given a high-fat diet. After 9 weeks, inguinal white adipose tissues were isolated for RNA extraction and hybridization on Affymetrix microarray. The RNA samples from three mice in each genotype were mixed for array analysis.
