The combination of rGO-FETs (reduced Graphene Oxide-Field Effect Transistor) and DNA oligonucleotide aptamers has proved to be a promising technical method to detect analytes with high sensitivity and specificity. Targeting the Amuc_1100 protein, we demonstrate the application and ability of the aptamer/FET combination to detect Akkermansia muciniphila, an intestinal symbiont that improves host metabolism. Amuc_1100, a protein highly expressed on the bacterial outer membrane that signals to Toll-like receptor 2 and, has been shown to partially replicate these beneficial effects. By developing, characterizing, and demonstrating the suitability of SELEX based anti-AMUC_1100 aptamer libraries for the construction of functional rGO-FET sensors for specific and sensitive Amuc_1100 quantification, we provide evidence that the evolution of focused polyclonal aptamer libraries for related targets can be used as a straightforward strategy. Revolution and innovation in sensing technology will undoubtedly further improve the diagnostic and monitoring process in biomedical technlogy.
