BIBW2992 is a small molecule tyrosine kinase inhibitor of EGFR and ERBB2 which has shown favourable clinical trial results in the treatment of head and neck squamous cell carcinoma (HNSCC). The Cetuximab and BIBW2992 sensitive HNSCC cell line JHU-029 has been treated with a fixed concentration of 0.1 mM ENU for 24 hours and then selected with BIBW2992 until drug resistant clones were ready to be picked and grown up as sub-clones of the parental cell line. These will have genes causally implicated in cancer sequenced to identify common point mutations in multiple independently derived drug resistant clones as a forward genetic screen for mechanisms of resistance to BIBW2992 in HNSCC.
