Rev1 has dCMP activity, which function as inserter at the abasic site. In addition to acting as an inserter, Rev1 plays an essential role in the insertion step functioning as a scaffold that recruits other inserter Y-family DNA polymerases.To investigate the mechanism of fork arrest mediated genome instability and its role in carcinogenesis, we generated mutants of rev1 gene in medaka fish (Oryzias latipes), and identified the alkylating agent-induced genomic instability in this mutant using a next-generation sequencer.
