Goals: to assess quality and mutation rate of a pooled plasmid library generated by error-prone PCR (epPCR). epPCR was used to mutate the coding sequence of synthetic human sequence (gene: PTGIR). The sequence was cloned into an E. coli plasmid (pcDNA5-HA, Life Technologies), isolated, and pooled for sequencing.
