The goal of this study is to analyze the impact of Srf overexpression on transcription during cardiomyocyte maturation in mouse Wildtype mice were injected with AAV9-cTNT-SRF-2A-GFP to overexpress Srf specifically in neonatal cardiomyocytes in mouse. AAV9-cTNT-GFP was used as control. Three biological replicates were analyzed in each group. In each experiment, cardiomyocytes were isolated by retrograde collagenase perfusion and purified by fluorescence activated cell sorting. Total RNA was extracted from FACS sorted cells. RNA-seq libraries were generated using SMART-seq and Nextera kits and were sequenced on a NextSeq500 sequencer. FASTQ Data were aligned by STAR and count matrix generated by FeatureCounts.
