The liver ductal organoid and the hepatocyte differentiation system provide an ex vivo system to investigate cholangiocyte-to-hepatocyte differentiation. However, as current gene manipulation method requires organoid dissociation into single cells and achieves low efficiency. Here we developed AAV-DJ vector as an effective delivery tool for liver bile ductal organoid. We transduced organoid with AAV-DJ-GFP and AAV-DJ-HNF4a to derive forced expression of GFP and HNF4a in ductal organoid. We found the forced expression of HNF4a could further induced the hepatocyte differentiation, compared with the GFP control. The sequencing data provide the a valuable resource to demonstrate AAV-DJ mediated forced expression of HNF4a could up-regulate hepatocyte marker genes globally.
