The main aim of this study was to investigate a direct alteration of the electrical properties of coupled cardiomyocytes by tissue-memory T cells from epicardial adipose tissue. Overall design: Epicardial adipose tissue was obtained from patients undergoing heart surgery. Immune cells were siolated and CD4 (CD3+ CD4+CD69+PD1+CCR7-CD45RO+) and CD8 (CD3+ CD4+CD69+PD1+CCR7-CDR5RO+) tissue-resident memory T cells were purified by FACS. As control, momory, non-TRM cells were also purified (CD3+ CD69-PD1-CCR7-CDR5RO+). Purified T cells were then co-cultures with iPSC-derived Atrial cardiomyocytes for 18 hours. Cells were washed and iPSC colected for RNA extraction.
