Pronounced early differentiation underlies zebra finch gonadal germ cell development.

We applied single-cell RNA sequencing to investigate inter-species differences in germ cell development between chicken and zebra finch (Taeniopygia castanotis, formerly Taeniopygia guttata castanotis), a Neoaves songbird species and a common model of vocal learning. Overall design: Early embryonic male and female gonads (HH28; n=2 per sex and species) were dissected and dissociated into a single-cell suspension using a 0.05% Trypsin solution. Library preparation was done using 10X Genomics 3' v3.1 kit. Sequencing was done using Illumina NovaSeq S2 flowcell.
